I can't reproduce.
thunderbird/claws mail installed, if I select Thunderbird in "Settings ->
Settings Manager -> Preferred Applications", thunderbird is run for mail, and
same for Claws Mail.

Which version of "exo" do you have ?
What is the output of ".config/xfce4/helpers.rc"
What is run wtih "exo-open --launch MailReader"
Does it work with xdg-open random at email.com ? Also check your
"~/.config/mimeapps.list" file
